چکیده

Hereditary factor XIII (FXIII) deficiency is a rare autosomal bleeding disorder which can cause life-threatening bleeding. Acquired be immune-mediated or due to increased consumption reduced synthesis. The most commonly used screening test insensitive, and widely quantitative assays have analytical limitations. present study sought validate Technofluor FXIII Activity, the first isopeptidase-based assay available on routine coagulation analyser, Ceveron s100. Linearity was evidenced throughout measuring range, with correlation coefficients of >0.99, variation for repeatability reproducibility were <5% <10%, respectively. A normally distributed reference range 47.0–135.5 IU/dL derived from 154 normal donors. Clinical samples Activity results between 0 167.0 assayed Berichrom® functional ammonia release assay, HemosIL™ antigen generating correlations 0.950 0.980, Experiments transglutaminase inhibitor showed that detect inhibition enzymatic activity. No interference exhibited by high levels haemolysis lipaemia, bilirubin evident at 18 mg/dL, level commensurate severe liver disease. rapid, accurate precise suitable diagnostic use fewer interferents than activity assays.

Measurement of factor XIII activity in plasma.

Coagulation factor XIII (FXIII) is converted by thrombin and Ca(2+) into an active transglutaminase (FXIIIa) in the final phase of coagulation cascade. Its main function is the mechanical stabilization of fibrin clot and its protection from fibrinolysis by cross-linking of fibrin chains and α(2)-plasmin inhibitor to fibrin. In non-substituted patients FXIII deficiency is a severe hemorrhagic di...

Automated homogeneous liposome-based assay system for total complement activity.

We developed an automated homogeneous immunoassay, based on immune lysis of dinitrophenyl (DNP)-labeled liposomes, for measuring total complement activity. Liposome lysis caused by complement activity was detected spectrophotometrically from entrapped glucose-6-phosphate dehydrogenase activity. Complement activity in human sera was quantified by comparison with a calibration curve. For ease of ...

Development and validation of an assay for urinary tissue factor activity.

BACKGROUND Activation of blood coagulation is a common complication of cancer and inflammation in both humans and experimental animals. Increased production of tissue factor--the principal initiator of the coagulation process--by endothelial cells, monocytes, and macrophages has been implicated in these conditions. AIM To investigate whether urinary tissue factor (uTF) might reflect the state...

Development and Validation of a Pilot Activity Load Index (PALI) based on NASA-TLX template

Abstract Introduction: Workload can be defined as the hypothetical construct that represents the cost incurred by a human operator to achieve a particular level of performance. Each job has specific needs and demands. The better measurement tool assessing that estimate the workload, it’s need to identify the requirements of a task, the circumstances under which it is performed, and the skills,...

Clinical significance of blood coagulation factor XIII activity in adult Henoch-Schönlein purpura.

BACKGROUND A correlation between decreased blood coagulation factor XIII activity and the severity of organ disorders in pediatric Henoch-Schönlein purpura (HSP) has been demonstrated, but possible correlations in adult HSP have not been thoroughly investigated. OBJECTIVES To investigate the association between factor XIII activity with varying clinical severities of HSP and the severity of o...
